About CARLOS SIMÓN FOUNDATION FOR RESEARCH IN WOMEN’S HEALTHAt Carlos Simon Foundation, we are offering an outstanding Predoctoral position in our research laboratory, based in Paterna, Valencia (Spain). The Carlos Simon Foundation for Women's Research is a non-profit organization founded and directed by Professor Carlos Simón, professor at the University of Valencia and renowned clinical researcher in the field of obstetrics and gynecology. Our main activity is biomedical research in the field of reproductive medicine and women’s health, focusing on clinical translation. As a multidisciplinary team of more than forty professionals, we are proud to be part of the INCLIVA Health Research Institute (IIS INCLIVA), accredited by the Carlos III Health Research Institute, Spain's leading national body for promoting health research.We are seeking one candidate to apply for a Predoctoral Contract in Health Research (PFIS) under the upcoming Call AES 2025 of the Carlos III Health Institute. Successful candidate will join the Maternal Fetal Communication research group at the Carlos Simon Foundation/INCLIVA Hospital Clínico de Valencia.Specifically, the project associated with the predoctoral researcher position is:Project PI24/01784 - Ovarian rejuvenation through partial cell reprogramming. Principal Investigators: Felipe Vilella Mitjana, PhD /Ana Monteagudo Sánchez. Project Theme:Women are born with a finite, non-renewable reserve of oocytes, the quantity and quality of which begin to decline considerably with age. This decrease is associated with a process called ovarian aging, in which different molecular and cellular mechanisms are involved. Cell reprogramming has been shown to enable the conversion of differentiated cells into pluripotent cells through the expression of Yamanaka factors. Expression of these factors over short intervals in aged cells restores juvenile epigenetic marks, inhibition of the senescence-associated secretory pathway (SASP), and reduction in the number of senescent cells. This process is called "partial reprogramming", it is a strategy that can promote broad cell rejuvenation effects and has been shown to restore aging-related cellular deficits in other tissues, we hypothesize that similar rejuvenation effects can also be achieved in ovarian cells. In this project, we aim to conduct a comprehensive study of single cells at the transcriptomic (snRNA-seq) and epigenetic (snATAC-seq and snmC-seq) level to understand what happens during ovarian aging. Subsequently, we will apply a partial reprogramming method based on CRISPRa technology in ovaries maintained ex vivo in a normothermic perfusion machine.
